I have an internal SSD in my HP ProDesk 4 Mini G1i that is not EXT4 formatted.
Therefore, I need to format this device to use it as my backup drive internally.
Is there a way to make this out of the Start OS, or do I need to boot a Linux Live system to achieve this?

Booting off a live Linux USB and using the disk utility will be the quickest and most straightforward way to go in order to format the drive to EXT4.
But you mention using this internal drive as a backup. This is something that is not supported at the moment by StartOS.
Backups can only be run to a network attached drive or to a USB connected drive. You can see these options by reading the Backups Documentation here:
